In the advanced lay-out editor there is the possibility to add 1/5th columns. But no option to add any other columns based on this size. I would like to use a 3/5th size column, is that possible?

No, at the moment not. I’ll mark this thread for Kriesi (feature request). Some other users requested the 2/5,2/5 & 4/5 layout too and maybe we’ll add it in the future.
