My goal is to have 1 image, that when clicked, opens a lightbox gallery with a number of related images and video.
I’ve come close. The Gallery element allows a style for “Big image only, other images accessed via lightbox”. However, the individual images within the gallery do not have a Custom Link field to insert a Youtube URL. *
On the other hand, I’ve been able to include video in lightbox galleries using a Masonry Gallery element, which does contain the Custom Link field allowing a Youtube URL. However, there does not appear to be an option to have 1 image launch all of the related images in the masonry gallery within a lightbox.
* As an aside, if the Gallery element is ultimately used. I would love to apply the image overlay (zoom effect) with the title on the big image… just like the Masonry Gallery options. If Gallery element is the way to go, eventually, I’d appreciate help with the CSS to make this happen.

While the Masonry Gallery & Gallery Element seem to be closely related, the differences that you describe would require quiet a bit of modifications to achieve the hybrid that you are looking for. Unfortunately this level of modifications is more than we can offer here.
I would recommend trying a plugin, such as: WP Video Lightbox or WonderPlugin Lightbox to name a couple that already seem to offer these functions.
